Detection of IL‑17/IL‑17A in Human PBMCs by Flow Cytometry. Human peripheral blood mononuclear cells (PBMCs) treated with PMA, Calcium Ionomycin, LPS, and Recombinant Human IL‑23 (Catalog # 1290-IL) to induce Th17 development overnight, re-stimulated with PMA, Calcium Ionomycin, and Monensin for 2 hours were stained with Mouse Anti-Human IL‑17/IL‑17A Alexa Fluor® 488‑conjugated Monoclonal Antibody (Catalog # IC3171G, filled histogram) or isotype control antibody (Catalog # IC002G, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005).
