| 描述 | The specific events which trigger the conversion of a primary malignant tumor into a more rapidly growing invasive neoplasm are largely unknown. One of the essential parameters for metastatic growth is the vascularization of the tumor, brought about by tumor angiogenesis factors. Recently, a new factor has been identified, designated angiostatin, which has been found to be an inhibitor of angiogenisis in a Lewis lung carcinoma animal model. Angiostatin is a 38 kDa proteolytic cleavage product of plasminogen, a 93 kDa protein. This factor has been shown to inhibit in vivo metastastic growth, regress primary tumors, as well as decrease the total number of vascular endothelial cells in vitro by inhibiting proliferation of these cells. The C9-1 clone recognizes the 93 kDa human plasminogen as well as the 38 kDa angiostatin fragment in western blot analysis. A recombinant protein containing the full length human angiostatin (361 amino acids, plus His Tag) was used as the immunogen. |
