| 背景 | 神经前体细胞表达发育下调蛋白8（NEDD8，Neural precursor expressed, developmentally down-regulated protein 8，在植物和酵母中又称Rub1，related to ubiquitin 1），是泛素样蛋白家族成员（1,2）。 NEDD8共价连接到靶蛋白，被称为neddylation，是类似于泛素化修饰的可逆的多步骤的过程。 NEDD8首先以前体形式合成，在羧基端含有一个延长肽，后被UCH-L3或NEDP1/DEN1水解蛋白切除产生成熟的NEDD8蛋白（3,4）。成熟NEDD8然后通过羧基端的甘氨酸残基，由APP-BP1/Uba3异源二聚体和Ubc12分别作为E1和E2类酶介导NEDD8共价连接到目标蛋白质的催化反应（5）。作为E3连接酶，Roc1/Rbx1，参与cullin蛋白的neddylation修饰（6）。蛋白去neddylation由细胞中的一系列催化酶介导发生，包括泛素特异性蛋白酶USP21、NEDP1/DEN1水解酶和COP9/signalosome（CSN）（7,8,9）。 与泛素修饰不同，NEDD8修饰系统只作用于少数底物蛋白，并不导致目标蛋白质降解。cullin蛋白的neddylation通过促进SCF (Skp1-Cullin-F-box) E3泛素连接酶复合体的形成和促进E2泛素中间体的招募来激活该复合体（10）。 VHL的NEDD8修饰虽然不是HIF1-α泛素化所必需，但参与纤连蛋白基质组装（11）。 MDM2依赖性的p53的neddylation抑制其转录活性（12）。 |
