| 背景 | TREML2 (TriggeringReceptor Expressed on Myeloidcells-like 2), alsol known as TLT2, is a 42-45 kDa member of the TREM family of immunemodulatory proteins. In mouse, it is a 306 amino acid (aa) type I transmembraneglycoprotein that contains a 246 aa extracellular domain (ECD). TREML2 isexpressed by CD5+ B-1 type B cells, marginal zone IgMhiIgDloB-2 type B cells, macrophages, microglia and neutrophils; it is absent from Tcells and monocytes (although human monocytes strongly express TREML2). While it is unclear what, or if, a proteinligand for TREML2 exists, it is known to bind phosphatidylserine and initiateefferocytosis (phagocytosis of apoptotic cells). TREML2 expression is promoted by TLR3ligation, and once expressed and "activated", it appears to potentiatedownstream effects that arise from independent GPCR activation, such asinflammatory mediator release and chemotaxis following chemokine binding to CXCreceptors. Over aa 25-270 (the ECD),mouse TREML2 shares 78% and 52% aa sequence identity with rat and human TREML2,respectively. With respect to mouseTREML2 ECD similarity to mouse TREM2 and TREML1 ECD, there is less than 20% aasequence identity for each. |
