| 背景 | TRAF6 is an adapter protein that links RANK with downstream signaling molecules, and plays a key role in RANKL mediated osteoclast differentiation. RANK contains a novel TRAF6-binding motif (T6DP) through which it binds to TRAF6 upon activation by RANKL. The TRAF6 inhibitory peptide contains a sequence from the T6DP motif of mouse RANK. The peptide binds to RANK, blocking TRAF6 and RANK interaction and thereby inhibiting osteoclast differentiation.
