| 描述 | Doxazosin is a non-selective α-adrenergic receptor antagonist with pKi values of 8.5, 9, and 8.4 for human α1A, α1B, and α1D adrenoceptors, respectively.1 Of the three α1 subtypes, α1A is seen as the primary regulator of smooth muscle tone in the bladder neck and prostate, α1B regulates blood pressure via arterial smooth muscle relaxation, and α1D is associated with contraction of the bladder muscle as well as sacral spinal cord innervation.2 Because of these roles for α1 adrenoceptors in mediating contraction of smooth muscle, doxazosin has been used to relax smooth muscles of the prostate in clinical studies designed to examine treatments that improve lower urinary tract symptoms associated with benign prostatic hyperplasia.2,3 |
