| 描述 | Leflunomide is a synthetic isoxazol derivative that regulates T lymphocyte progression through the cell cycle by selectively inhibiting de novo pyrimidine synthesis.1 As a prodrug, it is rapidly converted to its active open ring metabolite, A-771726 (Item No. 14404), which inhibits human lymphocyte proliferation with an IC50 value of 12.5 μM, in vitro.1 Leflunomide has been evaluated in phase II and III clinical trials for efficacy in slowing the disease progression of rheumatoid arthritis.1 It has also been used as an immunosuppressant agent to prevent rejection of transplant allografts and xenografts.2 |
