描述 | OP9细胞株源自新生的op/op 小鼠颅盖。因编码M-CSF的基因中的一个突变,它不能生成有功能的巨噬细胞克隆刺激因子(M-CSF)。M-CSF的存在对胚胎干细胞(ES)分化成血细胞而不是其他巨噬细胞有抑制功能。 OP9细胞可以用于与小鼠胚胎干细胞共培养以诱导胚胎干细胞分化成成红血球来源的、骨髓来源的和B细胞谱系的血细胞。与OP9共培养不需要外源的生长因子或复杂的胚胎结构。这个系统对研究造血细胞的发育和分化的分子机理有用。 |
