- 产品介绍
- background:
DNA polymerase lambda (pol Kappa), also designated DNA polymerase Kappa or Pol Beta2, is a low-fidelity polymerase which plays a role in both spontaneous and DNA damage-induced mutagenesis. Encoded by the POLL gene, pol Kappa is a member of the DNA polymerase type-X family. Pol Kappa extends primer-terminal mispairs opposite nondamaged DNA templates, suggesting that it may assist in extending mismatched base pairs during normal DNA replication. In addition, pol Kappa may play a role in the mutagenic bypass of T-T dimers. Proliferating cell nuclear antigen (PCNA), a protein essential to DNA replication, interacts with pol Kappa and thus influences the ability of pol Kappa to synthesize DNA.
Function:
Error-prone DNA polymerase specifically involved in DNA repair. Plays an important role in translesion synthesis, where the normal high-fidelity DNA polymerases cannot proceed and DNA synthesis stalls. Favors Hoogsteen base-pairing in the active site. Inserts the correct base with high-fidelity opposite an adenosine template. Exhibits low fidelity and efficiency opposite a thymidine template, where it will preferentially insert guanosine. May play a role in hypermutation of immunogobulin genes. Forms a Schiff base with 5-deoxyribose phosphate at abasic sites, but may not have lyase activity.
Subunit:
Interacts with REV1 (By similarity). Interacts with PCNA.
Subcellular Location:
Nucleus. Accumulates at replication forks after DNA damage.
Tissue Specificity:
Detected at low levels in testis, spleen, prostate and ovary. Detected at very low levels in kidney, colon, brain, heart, liver, lung, placenta, pancreas and peripheral blood leukocytes.
Similarity:
Belongs to the DNA polymerase type-Y family.
Contains 1 umuC domain.
