| 描述 | Thecsk gene encodes a protein kinase that phosphorylates several Src family kinases including p60[c-src], p59[fyn], p53/56[lyn], p56[lck], and p62[yes]. This phosphorylation occurs specifically at the carboxy-terminal tyrosine, Y527. Csk (c-srckinase) was identified based on its ability to phosphorylate exogenous Src in mammalian src-deficient cells. Establishment of a mouse csk- cell line revealed that phosphorylation of Y527 in Src was reduced 20-50%. This correlated with an increase of Src kinase activity, thus indicating Csk is a negative regulator of src.Csk-deficient cells also show an elevation in the kinase activity of p59[fyn] and p53/56[lyn]. These data suggest that Csk may be a universal negative regulator of Src family kinases in vivo. |
