| 描述 | Principle PRRSV is an RNA virus that is a member of the family Arteriviridae, order Nidovirales. PRRS viruses are classified into the European (EU/I) and the North American (NA/II) genotypes. PRRS virus infections can cause respiratory disease in piglets and reproductive failure in pregnant sows.ThepigtypePRRSV Ab and the pigtypePRRSV Ab OF are indirect ELISAs for detection of these viruses. The microtiter testplate is coated with recombinant PRRSV antigens, specific for NAand EU virus types. During sample incubation, antibodies specificfor PRRSV bind to the immobilized antigen. Unbound material isremoved by rinsing. Antibodies bound to the antigen are detectedby a horseradish peroxidase (HRP) conjugate. Unboundconjugate is removed by rinsing. A colorimetric reaction isinitiated by adding Substrate Solution and stopped after10 minutes. If antibodies specific for PRRSV are present in thesample, a blue color develops, which turns yellow after the addition of Stop Solution. The optical density (OD) is measured ina spectrophotometer at 450 nm. Measurement at a reference wavelength (620–650 nm) is optional. The OD value correlates to theconcentration of antibodies specific for the NA/EU strains ofPRRSV in the sample. Applications ThepigtypePRRSV Ab and pigtypePRRSV Ab OF are highly sensitive and specific solutions for detection of antibodies to PRRSV (NA and EU virus types) in serum/plasma and oral fluid samples from pigs, respectively.For further information and validation data on QIAGEN assays, contact QIAGEN Technical Service. |
