| 背景 | Lightning-Link is a one step antibody labeling kit, requiring just 30 seconds hands-on time. The antibody is covalently bonded to the label in a directional and controlled process at near-neutral pH.Label - PE/Atto594 is a completely new tandem conjugate. The PE is excited at 488nm and functions as an energy donor for the Atto594 fluorescent dye. Energy is transferred from the PE to the Atto594 dye via energy resonance transfer. The Atto594 emits the energy received from the PE in the form of long wavelength light at 627nm.Despite its simplicity, Lightning-Link is a very sophisticated conjugation system in which the antibody is directionally coupled to the label (and not to itself) in a controlled fashion, creating high quality conjugates. As the procedure is not interrupted by desalting steps, trial conjugates can be prepared with microgram quantities of protein and then scaled up with ease. Lightning-Link eliminates problems associated with scale up; hands-on time is essentially independent of process scale. Consistency from batch to batch is easy to achieve, and recoveries approach 100%. |
